ヒトサイトメガロウイルスgBのプロセシングにおけるcalnexinの役割

研究概要	ヒトサイトメガロウイルス糖蛋白質(gB)は105Kのnonglycosylatedの蛋白質として合成されるが合成と同時に小胞体内でN-linked oligosaccharideの付加を受け160Kのprecurso蛋白質となる。小胞体からゴルジ体へ輸送されるがゴルジ体でS-S結合でつながったgp55-116複合体へと内部切断をうける。我々はパルスチェイス実験により160KgB precursoが小胞体では膜結合シャペロンとして知られているcalnexinと一時的に結合していることを見いだした。その結合は合成直後が最大でhalf-time15分で解離する。又N-linked oligosaccharideはこの結合に重要である。さらに160KgB precursoは最初は還元型として合成され急速に酸化型へ変換する(half time18分)。両型ともcalnexinと結合できるが、還元型から酸化型への交換のkineticsはgB precursoとcalnexinの解離のkineticsと一致していた。
