新しいサイトカインIGIFによるアレルギー治療の試み

研究概要	IGIF(IFN-γ-inducing factor)は活性化マクロファージが産生する。我々の研究からIGIFは前駆体で存在し、LPSがマクロファージに作用すると、活性型のIGIFとして産生されること、そしてこの前駆体から活性型への変換に、ICE(IL-1βconverting enzyme)が関与することが明らかになった。また活性型のリコビナントIGIFをクローン化NK細胞に作用させるとFasLの発現が増強させることが明らかになった。更にIL-12とIGIFをT細胞あるいはB細胞に作用させると、T細胞だけでなくB細胞もIFNγ産生細胞に分化することも明らかになった。従って、抗CD40抗体とIL-4でB細胞を刺激してIgE産生を誘導する時にIL-12とIGIFを共存させた場合、B細胞自身がIFNγを産生する様になり、このIFNγがオートクラインまたはパラクラインにB細胞に作用してIgE産生を完全に抑制することも明らかになった。更にIL-12とIGIFを生体内に投与することで、寄生虫感染によって誘導されるIgE産生が抑制されることも明らかになった。その抑制機序は、IL-12とIGIFで刺激されると、in vivoでもT細胞とB細胞がIFNγ産生細胞に分化するからである。更にIL-12とIGIFはTh2細胞の誘導に必要と考えられるCD4^+NK1.1^+T細胞の数を著しく減少させることも今回の研究で明らかになった。我々は既にCD40L刺激がB細胞上にFas抗原を誘導することを明らかにしているが、IGIFがNK細胞やTh1細胞のFasLの発現を誘導することが今回の研究で明らかになった。この結果から、FasLを介したIgE産生抑制の系の存在も予測させるものとなった。今後はヒトのB細胞を用い、in vitroの方で同様の実験を行っていきたいと考えている。
