研究概要	本研究では腫瘍随伴神経症候群患者の細胞障害性T細胞の役割を明らかにすることを目的とした。患者T細胞と同一のMHCを有する標的細胞を得るため、患者の線維芽細胞の継代培養および末梢血リンパ球のEBウィルスによるトランスフォームを行った。これらの細胞をインターフェロンで刺激後、抗原としてリコンビナントYo蛋白を細胞内注入し、インターロイキンで刺激した同一患者のリンパ球を反応させた。リンパ球は、無処置の標的細胞あるいは牛血清アルブミンを注入した細胞に対しては反応が見られず、また、リコンビナントYo蛋白を注入したのみでリンパ球を加えなかったものでは細胞障害は認められなかったが、リコンビナントYo蛋白を注入した細胞はリンパ球を加えることによってその60%以上が消失した。今回用いた系では、細胞内に機械的に抗原を導入するため、生体内の条件とは異なる可能性があり、現在Yo蛋白をコードするcDNAを細胞内に導入したstable transformantを作製することにより、より生体に近い系でのT細胞機能の検討を行っている。一方、EBウィルスでトランスフォームしたB細胞ラインのHLA Class I,IIに結合しているプペチドをHLA分子から溶出し、溶出液から各ペプチドをHPLCにて分取した。今後各ペプチドのアミノ酸配列を解析し、その中から、使用頻度の高いペプチドモチーフを調べ、すでに判っているYo蛋白のアミノ酸配列と比較することで、T細胞およびB細胞の抗原決定基を推定する。
