病的心肥大関連転写因子TEF-1の心筋特異的活性制御機構

研究概要	転写因子TEF-1は、病的心肥大に際しβ-ミオシン重鎖など胎児型収縮蛋白遺伝子のプロモーターに特異的に結合してprotein kinase C(PKC)依存性に転写を活性化する。心筋細胞にはalternative splicingにより8種のTEF-1 isoform(α〜θ)が発現しており、その中にはPKCやcAMP-dependent protein kinase(PKA)により燐酸化されるisoformがある。TEF-1 isoformのうちα〜ζはβ-ミオシン重鎖の転写を活性化するが(転写活性化型isoform)、DNA結合能を欠くηとθは逆に転写を抑制する(転写抑制型isoform)。我々は以上の点を平成7年度までに明らかにしたが、平成8年度は以下の点を新たに解析した。(1)心筋肥大時に転写活性化型isoformが選択的に増加する可能性:培養心筋細胞の肥大(α_1-アドレナリン性受容体刺激による)に伴う各isoform mRNA量と各isoform間の比率の変化をRNase protection法により経時的に解析したが、大きな比率の変化は認められず、全体として発現が増大した。(2)プロモーター特異的転写制御:ミオシン重鎖(αおよびβ)と骨格筋型アクチンのプロモーターは転写活性化型isoformにより活性化されたが、ミオシン軽鎖プロモーターは影響を受けず、心房利尿ホルモンと脳利尿ホルモンのプロモーターは逆に抑制された。転写抑制型isoformはβ-ミオシン重鎖、心房及び脳利尿ホルモンのプロモーターのみを抑制した。TEF-1がこれらのプロモーターに結合する他の転写因子との複雑な相互作用を介して転写を制御することが明らかとなった。(3)ニ量体形成による転写制御:転写抑制型isoformは、転写活性化型isoformとニ量体を形成してそのDNA結合を阻害し、その結果β-ミオシン重鎖の転写を抑制することが証明された(dominant negative効果)。
