エンドセリン研究の動脈硬化症治療への展開

研究概要	私共は、既にエンドセリン-1が培養平滑筋細胞、培養メサンギウム細胞において遊離Ca^<2+>の上昇、DNA合成の促進をもたらすこと、さらにヒト冠状動脈由来の平滑筋細胞の遊走に対してエンドセリン-1単独では平滑筋細胞の遊走に影響を与えないものの、低濃度のPDGF-BB、アンジオテンシンII (Ang II)などの添加により相乗的に遊走反応を促進させることを報告した。酸化LDLが冠動脈硬化の発症・進展に関与していることが認められているが、今回、ヒト冠状動脈由来の平滑筋細胞の遊走に及ぼす酸化LDLの主要な構成リン脂質であるリゾフォスファチディルコリン(lyso-PC)の効果並びにエンドセリン-1との相互作用について検討した。lyso-PCは容量依存的に平滑筋細胞の遊走を促進し、その機序として内因性basic fibroblast growth factorの細胞内合成を促進させることが明らかにされた。また、エンドセリン-1はlyso-PCによる遊走反応を亢進させ、一方、エンドセリンA (ET_A)受容体拮抗薬であるBQ-123はこのエンドセリン-1の効果を抑制した。これらの結果はエンドセリン-1は酸化LDL内のlyso-PCとネットワークを形成して冠動脈硬化の発症・進展に関与していることを示唆しており、エンドセリン拮抗薬は、この意味で高コレステロール血症患者で認められる冠動脈硬化の予防薬・治療薬となる可能性も考えられる。しかし、この点はさらにin vivoにおいて検証すべきである。
