^<195m>Ptのトレーサー利用による各種白金化合物の制癌作用機構の解明

研究概要	4種類の制癌性白金化合物および三種類の非制癌性白金化合物を利用した標的量解析法により、CDDP,DWA2114R,CBDCA,254-S等のbifunctional白金錯体によるヒラ細胞の致死に対する標的がDNA分子であることが明らかにされた。また、致死に必要なDNA結合白金数がこれら制癌性白金化合物ではDNA構成ヌクレオチド約10^5当たり1分子となることも明らかとなった。すなわち、細胞致死効果をD_0で調べた際に見られたこれら薬剤間の作用の違いは薬剤が細胞中に侵入し標的分子と結合するに至る過程の違いに基づくものであり、結合白金当たりの細胞致死効率がいづれも大差がないことが判った。一方、TDDP等のmonofunctional錯体では標的量は10^3ヌクレオチド程度となり、結合白金当たり致死効率が上記錯体類に比べて著しく低くなることも明らかにされた。この様に、各薬剤の作用機構を明らかに出来たことにより、当初の目標は充分に達成されたと考えられる。次に興味あることは、白金化合物類と温熱との併用においてbifunctional chelating agent類よりもmonofunctional chelating agent類においてより高い協調作用が見られたことである。この事実から、温熱がmonofunctionalな結合にも強い相互作用を果たすことが結論された。最後に、^<109>Pdを用いて合成した標識シスパラジウムを利用した標識量の決定も進められている。シスパラジウムがシスプラチンに近い致死効率をもつことが明らかにされた。さらにまた、シスプラチンによって生じたDNA上の傷を特異的に認識し結合することが知られているテルビウムとシスプラチンとの併用作用についても標的量解析法が応用されている。それ自身では殆ど致死作用を持たない1.5mMのTbの存在により、シスプラチンによるヒラ細胞に対する平均致死濃度(D_0)が8.3μMから3.2μMとなり、著しい増感作用が確かめられた。これらの機構を明らかにするための検索も進められている。
