糖尿病性血管障害の解明-VEGFの遺伝子発現調節に関する研究-

研究概要	糖尿病合併症である網膜症、神経障害、更に動脈硬化症の予防と治療は、糖尿病臨床上重要な課題である。血管障害は血管内皮細胞および平滑筋細胞の機能が糖尿病の高血糖により変化を被ることが示唆され国際的に認識されている。そこで新しい血管新生因子であるVasculan Endothelial Growth Factor(VEGF)の(1)Human Studyおよび(2)Basic Studyにつき検討した。また糖尿病および:Vasculan biologyの真についても検討した。(3)。 (1)Human Study(ヒューマンスタディ) 筑波大学水泳部(8名)が、1886mの高地トレーニング(水泳トレーニング) (Kunming Chine)の前後(21日間)でのVEGF濃度、エリスロポエチン濃度を測定した。高地トレーニング19日でピークを認め23.0±5.3より46.0±14.6pgml^<-1>と有意な変化を認め、高地トレーニングにおける筋肉などでの酸素の効率化にVEGFが深く関わっていることを世界で始めて明らかにした。また論文として発表した。このことは糖尿病における運動についても多くのことを示唆する結果である。 (2)Basic Study(培養ヒト血管平滑筋細胞を用いた研究) ヒト血管平滑筋細胞のVEGF産生が低酸素刺激、コバルトおよび血管作動性物質であるET-1により亢進することを世界で始めて認め論文として報告した。その他(3)。
