骨髄細胞との細胞接着を介した骨芽細胞・破骨細胞の機能分化の制御:正常骨リモデリングならびにその病的破綻時における検討

研究概要

1.骨吸収と接着分子の関係
(1)マウス骨芽細胞のICAM-Iの発現はPTH、IL-1αで有意に、1,25(OH)2D3で軽度、prostaglandin E2では増加しなかった。(2)Raizeの系において、PTH、1,25(OH)2D3、IL-1αの投与による骨吸収増加はICAM-IおよびそのリガンドであるLFA-Iの抗体の投与で抑制されたが、prostaglandin E2による骨吸収はそれら抗体で抑制されなかった。(3)マウス骨髄細胞の培養系でPTH、IL-1α、1,25(OH)2D3による破骨細胞形成およびその活性促進はICAM-IおよびLFA-Iの抗体の投与で抑制された。一方、prostaglandin E2によるは破骨細胞形成およびその活性促進はそれら接着分子抗体で抑制されなかった。(結論)PTH、IL-1α、1,25(OH)2D3による破骨細胞形成および骨吸収促進作用に接着分子を介した細胞間接着が重要な役割をはたす。しかし、prostaglandin E2の骨吸収は接着分子を介した骨吸収に関与しないことが示唆された。
2.PTH、IL-1αによる接着分子を介した骨吸収における細胞内シグナル
(1)PTH、IL-1αによる胎児骨からの^<45>Caの遊出はPKA、PKC inhibitorで抑制された。(1)胎児骨からの^<45>Caの遊出はPKA、PKC activatorで促進し、その亢進はICAM-IおよびLFA-Iの抗体の投与で抑制された。(2)また、骨芽細胞上のICAM-Iの発現はPKA、PKC activatorで促進し、PTH、IL-1αによるICAM-Iの発現はPKA、PKC inhibitorで抑制された。したがって、PTH、IL-1αによる接着分子を介した骨吸収における細胞内シグナルはPKA、PKCの両者が関与する。
3.骨芽細胞の分化とICAM-Iの発現
PTH、IL-1α、1,25(OH)2D3は骨芽細胞の分化を促進する。これら因子による骨芽細胞の分化の結果、骨芽細胞のICAM-1の発現が増加した可能性がある。(1)IL-4は骨芽細胞の分化を抑制しするが、IL-4の投与は骨芽細胞上のICAM-1の発現を低下した。(2)マウス骨芽細胞(MC3T3E-1)のICAM-1の発現は培養後、骨芽細胞の分化とともに発現が上昇した。現在、骨芽細胞の分化マーカーとの対比を検討中である。
