発現クローニングによる血小板膜糖蛋白GPIIb-IIIa複合体の細胞内活性化因子の同定

研究概要	本研究では血小板膜糖蛋白(GP)IIb-IIIa複合体のリガンド親和性を変化させる未知の細胞内活性化因子を同定するための発現クローニングを行った。まずCMK細胞株をトロンボポエチン(TPO)で刺激して分化させた後にcDNA発現ライブラリーを作成した。発現ベクターとしてpBK-EFおよびpREPを用いた。発現ライブラリーをGPIIb-IIIa複合体をstableに発現するCHO細胞およびB細胞系株Namalwaに遺伝子導入しスクリーニングを行った。FITCラベルした精製フィブリノーゲンを加えたFlow Cytometerの系にて、より高親和性のGPIIb-IIIa複合体を発現する細胞を集め、そこからHirtの上清よりプラスミドを回収し、この系を繰り返す方法を行った。しかし数回のスクリーニングを繰り返しても、Flow Cytometerでの著名な蛍光の変化は無かったが、得られたそれぞれのクローンをピックアップしてシークエンスを行ったところ、そのうちの一つが、Yeast Two Hybrid SystemにてGPIIIaと結合する蛋白として報告されたβ3-Endonexinのalternative spliced formであることが判明した。FITCラベルした精製フィブリノーゲンでのassayではやはり感度に問題がり、代わりに活性型GPIIb-IIIa複合体に特異的な抗体PAC-1を用いて検討したところ蛍光の変化が観察された。別のスクリーニングの系として、骨髄巨核球から微量のmRNAを精製し、solid-phase RT-PCRにてライブラリーを作成し同様の発現クローニングを試みたが未知のクローンは得られなかった。GPIIb-IIIa複合体の活性化には単一の分子ではなくより複雑な経路が関与している可能性がありcomplementation cloningなどの系を試みる必要があると考えられる。
