内耳感覚細胞障害機構、回復機構の分子生物学的研究

研究概要

本年度は前年度に引き続き、まず、モルモットを利用して、耳石の変化と内リンパカルシウムイオンの恒常性維持機構について検討した。その結果、内リンパのカルシウムが急激に低下するような病的状態においては、最初に耳石よりカルシウムが流出することにより内リンパのカルシウム濃度を維持していることがわかった。この内リンパカルシウムの減少のための耳石の溶解はそれに引き続く再結晶化による多面型巨大耳石を生じさせ、さらに多面型の巨大耳石は移行型に変化する。この移行型の巨大耳石のカルシウムイオンの取り込みは低下しており、このことが回復過程での巨大耳石の消失を引き起こすと考えられた。これらのことから、耳石の量、形態の変化は内リンパのカルシウムイオンの恒常性の維持に密接に関係していると考えられた。また、前庭器における一酸化窒素(NO)について検討した結果、NOは内耳では主として構成型のNOSにより産生され、神経伝達や血流、内リンパの恒常性の維持に関与していることが明らかとなった。これに対して、誘導型のNOSにより産生されるNOは多量にかつ持続的に産生され、もう一つの主要なフリーラジカルである活性酸素と相互に作用し、より反応性に富むパーオキシナイトライトが生じ、これが内耳障害のメディエーターとして作用していることが明らかとなった。
これらの結果はSENDAI SYMPOSIUM'97,第6回日本耳科学会、第56回日本平衡神経科学会で報告するとともに、12編の論文にまとめられた。
