口腔粘膜の形態形成における各種成長因子の役割と相互作用の解析

研究概要

ラット・マウスの胎児(ラット:胎齢12、16日;マウス:胎齢15日)、生後(0、7、14、21日)の舌粘膜形態形成過程に関して、以下の点を明らかにした。
1. 走査電顕観察の結果から、ラット・マウスとも胎生中期に味覚性の茸状乳頭、有郭乳頭が非味覚性の糸状乳頭に先行して形成されることが明らかになった。糸状乳頭は出生直前から急速に形成され、味覚性・非味覚性の舌乳頭とも生後1週間位の間にほぼ成体のものに近似した形態を整えるに至る。
2. 光顕および透過電顕の観察結果から、ラット・マウスとも出生を境にして糸状乳頭部の舌粘膜上皮は急速に角化傾向を呈することが明らかになった。これは、胎生期と出生後の環境の違いに対する舌粘膜の適応的な変化と考えられる。
3. 胎生中期に出現する形成初期の茸状乳頭は、求心性神経線維の到達前に上皮の間葉側への陥入によって、上皮から離れた間葉内部にほぼ球形の原基として形成される。これらの原基は走査電顕観察で明らかにされたように、舌正中溝に沿った両側から順次外側にに向かって列を成して、舌背粘膜上皮に近接し、茸状乳頭の原基となる。求心性神経線維はその後に茸状乳頭の原基に到達し、原基内には味蕾が形成されて茸状乳頭となる。
4. 胎生中期の舌背粘膜上皮の上皮細胞は、単に非角化性であるだけでなく、生後動物とは異なり、立方細胞が上皮組織内に疎らに配列する構造を呈している。出生時にはすでに角化重層扁平上皮によって舌背粘膜上皮が構成されていることが光顕、透過電顕によって確認されたが、角化の進行と糸状乳頭の形成は生後急速に進展する。
5. EGF,FGE,TGF,NGFの免疫組織学的検討は光顕レベルでの観察が終了し、電顕レベルでの観察を継続中である。Cytokeratin subunitの免疫組織化学観察の結果については、現在結果の集計と解析をおこなっている。
