顎運動機能におけるヒスタミン神経系の役割

研究概要

今回取り上げた口腔顎顔面ジスキネジ-(orofacial dyskinesia)は難治性のものが多く、高齢化社会に基づくバ-キンソン病,痴呆をともなう精神病などの長期治療薬服用中(ハロペリドール,抗パーキンソン薬など)に多く見られる顎の異常運動である.この口腔顎顔面ジスキネジ-の実験モデルとして覚醒剤methamphetamineにより誘発される顎運動(oral stereotypy)が知られており,錐体外路系(とりわけ線条体)のドパミンとアセチルコリン神経系のバランスの異常が原因と考えられる.今回このモデルを用い,ヒスチジン処置によりヒスタミン神経系を賦活させるとmethamphetamineにより誘発される顎運動(oral stereotypy)が抑制された.一方,ヒスタミン受容体遮断薬メピラミン,ゾランチジン処置によりヒスタミン神経系を抑制させるとmethamphetamineにより誘発される顎運動が亢進した.また,神経化学的研究により,線条体のドパミンの遊離により顎運動が亢進されるが,ヒスタミンはドパミンより遅れて遊離して,ドパミンの過剰な刺激を緩衝しているという結果を得た。これらの結果より、ジスキネジ-の治療薬としてヒスタミンH3受容体拮抗薬が有効である可能性を示唆するとともに,ヒスタミン神経系は抑制系として顎運動に関与していることが判明した.
