研究概要	公共のデータベースに着実に集積しつつあるゲノム解析関連データをいかに創薬に役立てていくかは、薬学分野において喫緊の課題である。モデル生物の中でも最近その全ゲノム解析が完了した出芽酵母はヒト遺伝子とホモロジーのあるものが予想以上に多く、ヒト遺伝子の機能解析の上で重要なツールとなるであろうことが再認識された。本研究では出芽酵母を用いて、情報科学に基づく機能的モチーフによる遺伝子の分類を出発点とした遺伝子機能の解析系を出芽酵母をモデルに用いて確立することをめざして、研究を行った。本研究により達成されたことは以下の各項である。 (1)効率の良い出芽酵母遺伝子破壊系の確立:遺伝子破壊の際に用いるマーカー遺伝子を工夫することにより、遺伝子破壊株が得られる確率が従来の数%から30%にまで改善された。 (2)重要な創薬ターゲットとなりうるレセプター遺伝子・情報伝達に関与する遺伝子の遺伝子破壊株の性状解析手法の確立:酵母細胞の代謝活性をリアルタイムに測定することができれば、レセプターとリガンドの相互作用の詳細な解析が可能になる。microphysiometerを用いることにより、このような測定が可能となった。 (3)酵母遺伝子発現系の確率:増殖に必須の遺伝子のクローニング・大腸菌及び酵母での発現系を確立した。ヒスチジンtagをつけた形で発現させ得るようになったので、遺伝子産物のアフィニティ精製も可能となった。 (4)以上の成果に加え、実際に遺伝子破壊実験を行い、新規増殖必須遺伝子の同定とその遺伝生化学的な解析をも進めた。
