硫酸化環状オリゴ糖による内因性組織修復因子の活性化を介した難治性潰瘍の治癒促進

研究概要

消化管粘膜損傷時に動員される内因性組織修復因子の安定性および病巣到達性を高め、潰瘍の治癒促進を企図する新しいタイプの潰瘍治療薬の開発を目指して、各種硫酸化シクロデキストリンおよびその難水溶性アルミニウム塩を調製し、それら誘導体の潰瘍治癒促進作用を比較検討した結果、以下の新知見を得た。
1)各種硫酸化シクロデキストリンの難水溶性アルミニウム塩は、代表的な内因性組織修復因子である塩基性線維芽細胞増殖因子(bFGF)に対して強い親和性を示し、シクロデキストリン空洞サイズや置換基の種類よりも、硫酸基の置換度に依存して親和性が変化した。
2)bFGFと硫酸化シクロデキストリンの難水溶性アルミニウム塩の吸着体は水中で長時間安定に存在したが、等張リン酸緩衝液(pH7.4,37°C)中では吸着体からbFGFが徐々に遊離した。その際、脱着したbFGFは未処理bFGFと同等の細胞増殖活性を保持していた。
3)硫酸化シクロデキストリンのアルミニウム塩は、消化酵素(α一キモトリプシンやペプシンなど)によるbFGFの分解を顕著に抑制した。その安定化効果はデキルトラン硫酸やショ糖硫酸エステルアルミニウム塩などよりも強力であった。
前年度の研究成果を踏まえて、本年度に得られた実験結果を総合評価すると、硫酸化シクロデキストリンの難水溶性アルミニウム塩は内因性組織修復因子の安定化を介して難治性潰瘍の治癒を促進する新規抗潰瘍薬として有効利用が期待される。
