研究概要	GPI-アンカー型蛋質はglycosyl phosphatidylinositolを介して細胞膜に結合したユニークな構造をもつ蛋白質群である。GPI-アンカー型蛋白質を抗体でクロスリンクすると、細胞内Ca^<2+>濃度の上昇、細胞増殖が誘導されることから、これらの分子がシグナル伝達機能をもつと考えられている。しかしながら、細胞内ドメインを持たないこれらの分子が、細胞質側にどのようにシグナルを伝えるのかのかは不明である。本研究では、GPI-アンカー型補体制御因子CD59に焦点をあて、これをモデルとしGPI-アンカー型蛋白質のシグナル伝達機構について明らかにすることを目的とする。 GPI-アンカー型蛋白質による細胞の活性化は、特異抗体を作用後、さらに2次抗体でクロスリンクするという方法が一般に用いられている。このような方法では、Fcを介した活性化の関与を否定できない.また、細胞の活性化を免疫沈降などで検出する際には、抗体の存在が測定上の障害となる。そこで、シグナル伝達を誘導しうるligandを見つけ、これによる測定法を開発することが重要である。CD59は、C9に結合し、C9分子の重合を阻害することにより補体系を抑制する。同様にC9分子が、C59の細胞活性化の際のligandとなりうる可能性がある。本年度は、種々のC9ペプチドを合成し、これらの効果を「細胞内Ca濃度の上昇」を指標として測定した。その結果、ペプチド2(407-436)がJurkat細胞で、細胞内Ca^<2+>を特異的に上昇させることが明らかとなった。至適濃度は50μMであった。細胞外のCa^<2+>を除くと、細胞内Ca濃度の上昇は50%程度抑制されたことから、Caは細胞外、細胞内、両者に由来すると考えられる。今後、チロシンキナーゼの活性化、細胞増殖などへの効果についても検討しCD59シグナル伝達が誘導される系を確立したいと考えている。
