ムラサキツユクサ雄蕊毛による異なる環境変異原間の相乗効果の研究

研究概要

環境制御した培養液循環栽培装置内でムラサキツユクサBNL4430株(青/ピンクのヘテロ株)の若い花序をもつ根つきshootsを大量に育て、雄蕊毛におけるピンク色体細胞突然変異誘発頻度を指標として、異なる変異原間の相乗効果の研究を継続した。
これまでに、単一作用型のアルキル化剤であるメチルメタンスルホン酸(MMS)、硫酸ジメチル(DMS)、エチルメタンスルホン酸(EMS)、N-メチル-Nニトロソウレア(MNU)、N-エチル-Nニトロソウレア(ENU)のうち、MNUはX線と加算効果しか示さないが、他はすべてX線と明白な相乗効果を示し、X線との相乗効果とその現れ方がこれらアルキル化剤のSwain-Scott基質係数(s)、すなわちMMS:0.88、DMS:0.86、EMS:0.67、MNU:0.42、ENU:0.26と必ずしも一致しないことが判明していた。そこで、やはり単一作用型のアルキル化剤である硫酸ジエチル(DES)とX線との相乗効果を調査したところ、明白な相乗効果が見られたものの、その現れ方は、やはりs値とは一致しなかった。また、MMSとEMSの間では明白な相乗効果が見られたのに対して、MNUとEMSは単に加算効果しか示さなかった。
Promutagenであるマレイン酸ヒドラジド(MH)、o-フェニレンジアミン(PDA)、N-ニトロソジメチルアミン(DMN)のいずれもが、X線照射後処理では明白な相乗効果が現れるのに対して、処理後X線照射では相殺効果が見られ、ペルオキシダーゼ活性が、前者では高まるのに対して、後者では抑えられ、この酵素がこれらpromutagenの活性化に関与していることが判明していた。ただし、DMN、PDA、MH処理中X線照射は、それぞれ相乗、加算、相殺効果を示した。promutagenでありかつ二作用型のアルキル化剤である1,2-ジブロモエタン(EDB)処理中のX線照射は明白な相乗効果を示したが、MHとEMSは常に相殺効果を示した。
