細胞分化因子としてのインスリン作用の情報伝達系の解析

研究概要	3T3-L1繊維芽細胞はインスリン、デキサメサゾンおよびイソブチルメチルキサンチン(IBMX)の3種の混合液(分化誘導薬)を添加して培養することにより、脂肪細胞へ分化する。これら3者の薬物は各々脂肪細胞への分化に対して重要な役割を担っているがその中でも、インスリンが重要な役割を担っていることが分かった。すなわち、脂肪細胞特有の性質である脂肪合成酵素系の活性化による中性脂肪の蓄積およびグルコーストランスポーター4型の発現によるグルコース取り込みの亢進はインスリンの作用によるものである。したがって、3T3-L1繊維芽細胞の脂肪細胞への分化に対するインスリンの情報伝達系を中心に検討した結果、以下のことが明らかとなった。(1)インスリンによる脂肪酸合成の亢進は抑制性のGTP結合蛋白質Giの機能を消失させた百日咳毒素処理によって著しく抑制された。(2)インスリンはイノシトールグリカン特異的ホスホリパーゼC(P1-PLC)の活性化を引き起こしたが、百日咳毒素処理はこのPI-PLCの活性化を抑制した。(3)PI-PLCの活性化はdiacylglycerolの生成を伴い、C kinaseの活性化を引き起こす。そこで、直接C kinaseを活性化させるPMAを添加すると、脂肪細胞への分化は抑制され、C kinaseを消失させるPMAの長時間処理は逆に分化を増強させた。(4)各種の細胞において、増殖や分化の過程にプロトオンコジーンの発現が起こることが報告されている。3T3-L1繊維芽細胞に分化誘導薬を添加すると、c fos発現が認められ、百日咳毒素処理はこの発現を完全に阻害した。分化誘導薬中のIBMXが3T3-L1繊維芽細胞の増殖を抑えていたことから、百日咳毒素処理は分化の過程に必須なc fos発現を阻害したと推定した。以上の結果から、インスリンによる3T3-L1繊維芽細胞の脂肪細胞への分化の過程に百日咳毒素感受性のGi蛋白質が介在し、PI-PLCの活性化→ →c fos発現→ →脂肪細胞への分化が起こるものと推定した。一方、C kinaseの活性化は脂肪細胞への分化を負に調節していると結論した。
