GM-CSFによる細胞増殖制御機構の研究

研究概要	本年度は、研究の中心となるin vitro系の開発に主眼をおき、成功した。プロB細胞であるBA/F3細胞より細胞膜、細胞質画分をそれぞれ調整し、in vitroでATP再生系とともに反応すると、GM-CSFと、GM-CSFレセプターに依存してSTAT5の活性化が誘導された。この活性化は膜、細胞膜画分の両者とATPが必須であり、抗リン酸化チロシン抗体で阻害される。従ってレセプターの活性化に続いてチロシンキナーゼが活性化され、STAT5の活性に至ることが予測された。実際にin vitroで活性化された反応液からJAK2を免疫沈降するとチロシンリン酸化がGM-CSFによって誘導されていることが明らかになった。我々はこれまでにin vitroのGM-CSFレセプターの変異体の研究からGM-CSFレセプターの下流には増殖/c-mycの誘導に至る経路、MAPKを活性化してc-fos活性化に至る経路がSTATの活性化の経路以外に存在することを明らかにした。in vitro系の検討の結果によれば、Shc、PTPIDなどMAPKの上流のシグナル伝達経路が活性化される。増殖の機構を検討為にGM-CSFレセプターの膜貫通領域の直下のサイトカインスーパーファミリーで保存されているBOX 1/BOX 2のGST融合タンパク質を使い、相互作用する分子を検索した。これまでに構成的、誘導的にこの領域に結合する複数の分子の存在が明らかになった。この中で、Srcキナーゼの一員であるLyn、HcKを結合することが確認された。現在これらの相互作用と共に他の分子の同定及び遺伝子の単離を進めている。
