ヘミデスモソーム膜タンパク質BP180の分子形態と機能の解析

研究概要

本研究は細胞・基質間接着装置ヘミデスモソームに存在する分子、特にコラーゲン様膜タンパク質BP180の生化学的性質を調べ、リガンドを同定し、他の構成成分との相互作用を調べることにより接着構造に果たす機能を明らかにすることを目的として行った。具体的には次のような成果を得た。
1 ウシ乳腺由来のBMGE+H細胞の可溶性画分からモノロ-ナル抗体を用いたアフィニテ-カラム法で十分量の可溶性180KD分子を精製することに成功した。
2 BP180分子の細胞外領域は分断されたコラーゲンドメインよりなるので、この分子が実際に3量体を形成し、コラーゲン型3重らせん構造を形成していることを架橋剤を用いて生化学的に明らかにした。
3 単離したBP180分子を低角度回転蒸着法により、電子顕微鏡で分子形態を直接観察することに成功し、分子は球状の頭部、棒状の中央部、柔軟な尾部からなる八分音符形をしている事を明らかにした。
4 BP180の細胞質側結合分子を探るために、BP180の細胞外部分に対するモノクローナル抗体を用いて、BP180分子を結合させたアフィニテ-カラムをつくり、可溶化した細胞質成分から相互作用するタンパク質を探した結果、230kDのタンパク質が弱いながらもBP180に結合するとのデータを得た。
5 共同研究により、以下の成果をあげた。
1)水疱症にはBP180分子が遺伝的に欠失している型が存在することを明かにした。
2)水疱症に筋ジストロフィーを併発する型では、ヘミデスモソームタンパク質HD1が欠失していることを証明した。
