研究概要	出芽酵母の出芽過程は細胞の極性を形成する過程であり、出芽酵母は細胞極性形成の分子機構の研究において格好なモデル生物となっている。一方、私共を含むいくつかのグループは、Rhoファミリーの低分子量G蛋白質が、カスケード的に働いて酵母の出芽過程を制御していることを明らかにしている。本研究では、この低分子量G蛋白質カスケードの分子機構について解析を行い、以下の成果を得た。低分子量G蛋白質カスケードを明らかにするためには、低分子量G蛋白質の活性制御蛋白質や標的蛋白質を単離することが不可欠である。私は、本研究において、RHO1のGDP/GTP交換反応を促進してRHO1を活性化する蛋白質としてROM1とROM2を発見した。また、RHO1と直接相互作用する全く新しいタイプの活性制御蛋白質としてROM7/BEM4を発見した。一方、私は、RHO1の標的蛋白質が、プロテインキナーゼCのホモログであるPKC1とグルカン合成酵素、BNI1/BNR1であることを明らかにした。さらに、私は、RHO1が、PKC1を介して遺伝子の発現を、グルカン合成酵素を介して細胞壁の合成を、BNI1/BNR1を介してアクチン細胞骨格の再編成を制御することにより、酵母の出芽過程を統合的に制御していることを明らかにした。特に、RHO1は、BNI1/BNR1を介してアクチン結合蛋白質であるプリフィリンに作用して、アクチンの重合過程を制御していることを明らかにした。以上の成果を得たことにより、本研究の目的は十分に達成できた。
