両生類精巣における成長因子の探索と精子分化に対するその生理活性

研究概要	精子分化過程における体細胞分裂を行う精原細胞から減数分裂を行う精母細胞への分化の機構という問題を研究するために、我々は、イモリの器官培養系や細胞培養系において、FSHの処理によってセルトリ細胞から出される増殖因子(あるいは減数分裂誘導因子)を同定し、そのシグナルを受けて精原細胞が増殖し、減数分裂が開始するメカニズムを解明することを目的とした。 (1)器官培養における種々の増殖因子の影響精原細胞ステージの精巣断片を器官培養し、種々のヒトリコンビナント増殖因子を加え、1週間後にBrdUを取り込ませて精原細胞の増殖活性を調べた。コントロールでは1週間後約5%まで低下したが、FSHを加えた培地では、約35%と高い値を保っていた。コントロール培地にヒトIGF-Iを50ng/ml加えたところ、30%弱の増殖促進活性が見られた。2週間後分化率を調べると、FSH培地では約45%のシフトが第一精母細胞へ分化した。IGF-IはFSHよりは低いものの20%弱のシフトで分化した。 (2)イモリFSHリセプターcDNAの単離精原細胞ステージの精巣断片から調整したRNAを鋳型として、哺乳類のFSHリセプターの細胞外ドメインの2カ所で設計したプライマーを用い、RT-PCRを行ったところ、831bpから成るcDNA cloneが得られた。ヒトのFSHリセプターと塩基レベルで約65%、アミノ酸レベルで約58%のホモロジーが見られた。
