脳神経系特異的新規チロシンキナーゼ型レセプター"sky"の機能解析

研究概要	申請者らは、水野、大橋らとともに世界に先駆けて、最近レセプター型チロシンキナーゼをクローニングし、skyと名付け、このhuman skyに続きさらにrat skyをクローニングした・Skyはその構造上、多くの栄養因子レセプターの持つチロシンキナーゼドメインを持つほか、細胞外に神経栄養因子のレセプターに存在するIg-likeドメインと神経接着因子の持つFibronectinIII-likeドメインを持つ非常にユニークなレセプターである。Skyは脳神経系に特異的に存在している。その構造上のユニークな特性と合わせ、Skyは、神経の栄養・ガイダンス等神経組織に重要な新しい機能を持ったレセプターと考え研究を進めている。 Skyには同じ構造を持つチロシンキナーゼ型レセプターである他の2つのファミリーメンバー(Axlおよびc-Mer)の存在が報告されている。それらのリガンドとしてはGas6およびProtein Sが報告されているが、レセプターリガンド対応に関しては、種により異なる結果が報告されている。 (1)そこで全て1つの種(ラット)で解析すべくSkyおよびそのリガンドファミリーの部分遺伝子をPCR法を用いて獲得した。ラットaxl遺伝子は報告がなかったがこの全長をクローニングし、juxtamenbraneドメインの長さの異なる2種のバリアントを認めた。これらを用いてmRNAの発現分布をRNAseプロテクションアッセイ法を用い、血管内皮細胞の増殖と重要とされるgas6や血液中に豊富なprotein S、またsky,axlおよびc-mer mRNAの脳神経系、さらに各種cell line、初代培養海馬神経細胞、アストロサイトやミクログリアにおける発現の解析を進めている。またラットSkyおよびラットAxlのペプチド抗体を作製、アフィニティー精製し、これを用いin situハイブリダイゼーション法による解析に加え免疫細胞、組織染色法を用い解析を進めている。 (2)Skyが高発現する海馬の初代培養ニューロンを用い、そのリガンドによる神経生存作用を評価、Skyが神経栄養因子レセプターとして機能できるか否か、またそのメカニズムを解析中である。 (3)Xenopusの系で解析するためxenopus sky family遺伝子をクローニング、新規ファミリー遺伝子の可能性を含め解析中である。
