脳神経系特異的新規チロシンキナーゼ型レセプター“sky"の機能解析

研究概要

Skyは、私たちのグループにより1994年にクローニングされた新規チロシンキナーゼ型レセプターである。Skyはその構造と発現分布から神経系において重要な機能をもつと考えられるが、これまでSkyの神経系における機能は全く不明であった。本研究では、ラットGas6が培養海馬ニューロンに対する神経生存促進因子、すなわち新しい神経栄養因子として機能することを発見した。このことは、Gas6/Skyファミリーシステムの新規神経栄養システムとしての発見である。
またSkyファミリー分子ラットAxlを分子クローニングし、この配列をもとにラットSkyおよびAxlに対する抗体を作製し組織・細胞分布を解析すると共に、Skyのファミリーレセプター、リガンドmRNAの発現分布、発達過程における発現制御、更には神経損傷時における生理的発現制御(カイニン酸モデルと座骨神経損傷モデル)を解析した。発生過程および神経損傷モデルにおける協調したGas6/Skyファミリー分子の神経系における発現制御は、Gas6/Skyファミリーの機能が生理的に重要であることを裏づけている。更にアフリカツメガエルの系を用いた機能解析を目指し、アフリカツメガエルよりSkyファミリー遺伝子“Xkisy"をクローニングした。Xksyはその発生過程で神経系に発現が片寄っていたことから、神経発生に重要な機能をもつことが示唆された。私たちは、Gas6の海馬ニューロンに対する作用に加え、他の複数のニューロンがGas6に応答することを見い出している。また、Gas6/Skyファミリーシステムのもつ神経発生過程や神経損傷時における神経ガイダンス能の可能性についても解析中である。今回明らかとなった新規神経栄養システム(Gas6/Skyファミリーシステム)の解析が今後更に進み(遺伝子変異体、トランスジェニック、ノックアウトマウスやアフリカツメガエルの系等を用い)、神経ネットワークの構築・維持・再構築のメカニズム解明の一助となることが期待される。
