ラン藻における光誘導性遺伝子発現制御機構の解析

研究概要

本研究では明暗の光変化に応答した遺伝子発現の根本的な転写制御システムを理解する手始めとして、ラン藻M.aeruginosa K-81株をモデル生物として、本菌における光合成遺伝子群の一つであるpsbA2の光誘導性転写発現の解析を行った。まずpsbA2遺伝子のプロモーター欠失クローンを作製し、大腸菌とラン藻内でプロモーター活性を測定した。その結果、psbA2遺伝子の転写に必須な領域(-38〜+14)と光誘導に関与する領域(-80〜+14、+15〜+46)を同定した。同定された光誘導に関与する領域の塩基配列を他種ラン藻や高等植物のpsbA2遺伝子群の5'-上流領域塩基配列と比較したところ、約10bpのAT-richな共通配列を見出した。更にpsbA2遺伝子のプロモーター領域で、新規のDNA湾曲構造(CIT:changeable bending-center sites of an intrinsic curvature under temperature conditions)を発見した。
次にpsbA2遺伝子の発現に関与する転写調節蛋白質を解析する一環として、in vitro(試験管内)転写実験系を確立させた。K-81株より主要シグマ因子(σA1)を含んだRNAポリメラーゼホロ酵素を部分精製し、鋳型psbA2プロモーターDNA断片からの特異的な転写産物を(単細胞性ラン藻の無細胞抽出画分を使用した系では初めて)確認するとともに、-38〜+14が転写の最小必須領域(minimal cis-element)であることを証明した。
