副腎皮質ホルモンによる視床下部一酸化炭素合成酵素遺伝子発現調節についての検討

研究概要	一酸化窒素(NO)は生体内では、L-アルギニンを基質としてNO合成酵素(NOS)により産生される。神経型NOSは、自律神経系と内分泌系の統合部位である視床下部室傍核(PVN)および視策上核(SON)に多く発現している。一方、NOと同じモノオキシドであるCOもまた脳内で逆行性伝達物質として働いている可能性が報告されている。COの合成酵素はヘムをビリベルジンに変換するヘムオキシゲナーゼ(HO)-1,-2である。HO-2は脳内に高濃度で存在し、視床下部にも発現している。今回我々は、In situ hybridization法を用いて、ラット視床下部における神経型NOSおよびHO-2遺伝子発現の変化について、高張食塩水負荷後または副腎摘除後で検討した。実験にはウイスター系雄ラット(200-250g)を用い、2%高張食塩水を給水ボトルで与えた群(n=6)と水道水を与えたコントロール群(n=6)および両側副腎摘除群(n=6)とSham手術群(n=6)の4群を作成した。各群ともに5日後に断頭して、脳を取り出しドライアイス上で凍結した。クリオスタットを用いて12μmの凍結切片を作成し、固定、脱水および脱脂後、RI(^<35>S)標識した合成オリゴヌクレオチドプローブ(神経型NOS、HO-2およびCRH)で一晩ハイブリダイゼーションした。洗浄・乾燥後、高感度フイルムに露光した。その結果、神経型NOSmRNAは、高張食塩水負荷群で、PVN・SONにおいて著明に増加したが、副腎摘除群では変化なかった。HO-2mRNAは、高張食塩水負荷群および副腎摘除群でPVNにおいて増加傾向が見られた。CRHmRNAは、副腎摘除群のPVNの小細胞群で著明に増加した。したがって、視床下部においてNOは、バゾプレッシン系を介した体液調節に、COは浸透圧調節および視床下部-下垂体-副腎系に関与していることが示唆された。
