標的遺伝子組換えによるグリア細胞特異的内向き整流カリウムチャネルの解析

研究概要

1)K_<AB>-2チャネルゲノム遺伝子の構造解析:マウスゲノムライブラリーよりK_<AB>-2ゲノム遺伝子を単離し、その構造解析を行った。K_<AB>-2遺伝子はエクソン1及び2からなり、蛋白コーディング領域にはイントロンが存在しないことを明らかにした。現在、エクソン1上流のプロモーター領域の解析、さらにK_<AB>-2遺伝子の染色体マッピングを行っている。
2)K_<AB>-2チャネルを発現しないノックアウトマウスの作成:上記遺伝子を利用してターゲティングベクターを作製した。今後は、このベクターをES細胞に導入し相同組み換えの生じている細胞をサザングロット法により選別する。得られた細胞を宿主胚盤胞に注入し、偽妊娠雌マウスの子宮内に移植することによりキメラマウスを得る。これらのキメラマウスを使い、相同組み換えが生じているノックアウトマウスを得る。
