情報伝達を担うリン酸化酵素群の細胞内における活性測定法の開発と応用

研究概要	プロテインキナーゼC(PKC)とプロテインキナーゼA(PKA)の細胞内活性測定法の確立細胞骨格タンパク質、ビメンチンのSer33とSer46はそれぞれPCKとPKAによって特異的にリン酸化を受ける部位である。従って、細胞内のビメンチンSer33とSer46のリン酸化をモニターすることにより細胞内のPKCとPKAの活性を測定することができる。申請者らは、今回Ser33のリン酸化を認識する抗体を作製した。Ser46リン酸化認識する抗体は現在作成中でらあるが、PKAによってリン酸化を受けるSer46以外の部位であるSer6とSer38のリン酸化を認識する抗体の作製に成功した。現在これらの抗体を用いて培養細胞のPKC、PKAを実際に活性化させ、酵素活性がモニターできることの確認を行っている。培養グリア細胞内におけるCaMキナーゼII(CaMKII)活性の空間分布の検討申請者らはビメンチンのSer82のリン酸化をモニターするモノクローナル抗体を作製し細胞内のCaMKIIの活性を測定可視化する方法を確立している。今回、培養グリア細胞でこの方法を用いてCaMKIIが細胞内に限局して活性化することを世界に先駆けて見出した。またこの方法とカルシウム顕微鏡とを組み合わせることで、カルシウムシグナルの細胞内分布が細胞内情報として、下流に存在するCaMKIIへと伝えられることを証明した。個体レベルにおけるニューロン、グリアにおけるCaMKIIの活性化の検討申請者らは上記の方法を用い、培養細胞のみならず個体、組織レベルにおいてもニューロン、グリア細胞内でCaMKIIが活性化することを確認した。現在、個体発生に伴うニューロン、グリアの分裂、移動、分化とCaMKIIの活性化との関係を検索中である。
