研究概要	ニューロン、グリアへの遺伝子導入によるパーキンソン病治療の基礎的検討神経細胞あるいはその周囲に存在するグリア細胞への有効な遺伝子導入法が確立されれば、難治性の中枢神経疾患の治療への道が開けると考えられる。我々はセンダイウイルスの細胞融合能を利用したHVJーリポソーム法により、ラット小脳顆粒細胞(初代培養神経細胞)の細胞核内にアンチセンスオリゴヌクレオチドやβ-galactosidase遺伝子を高効率での導入に成功し、しかも長期にわたって発現させることができた。また生体においてもマイクロインジェクション法とHVJ-リポソーム法の併用でラット脳内に遺伝子を導入し、その産物の4週間以上にわたる機能発現を確認した。さらに、現在進行中の実験として、強力な神経再生因子としての作用を有する肝細胞増殖因子(HGF)、及びドーパミン受容体(D2)遺伝子を培養小脳顆粒細胞、培養ヒトアストロサイトに導入することで、長期のドーパミン産生とその作用の安定性を検討している。具体的には、CMWプロモーターを持つプラスミドベクターに挿入したHGF及びドーパミン受容体(D2)遺伝子を初代培養神経細胞にHVJ-リポソーム法を用いて導入し、mRNAレベルでの遺伝子発現をノーザンブロッティング法およびRT-PCR法にて経時的に確認している。また産物の活性測定についても検討を行っている。本研究では遺伝子治療の基礎的検討を行っているが、今後パーキンソン病の発症機構の詳細が解明されれば、さらに有効な根治的遺伝子治療が可能であると考えられる。その際、本研究で得られた知見が大いに役立つと考えられる。
