ヒト椎間板マトリックスメタロプロテアーゼの産生調節機構と椎間板変性機序と制御

研究概要

静水圧のヒト椎間板基質合成能(プロテオグリカン、プロテイン合成能)および椎間板基質分解能(基質分解酵素Matrix metalloproteinase-3(MMP-3)、およびその抑制物質Tissue inhibitor metalloproteinases-1(TIMP-1)産生能)におよぼす影響を明らかにした。椎間板髄核においては0.3MPa(3気圧)の生理的静水圧下では大気圧下に比べてMMP-3、TIMP-1産生能は変化を示さず、プロテオグリカン、プロテイン合成能の促進を認めた。逆に、3MPa(30気圧)の高圧負荷群においては合成系に変化を及ぼさないものの、MMP-3産生亢進、TIMP-1産生低下を引き起こした。線維輪においても同様の傾向であったが、特に高圧負荷群においては合成系の抑制、および分解系の亢進を認めた。
静水圧は椎間板基質合成能、および分解能にその圧力の程度により促進的、または抑制的効果を持つことが明らかとなり、種々の成長因子、サイトカインのように重要な細胞機能調節因子と考えることが出来る。適度な力学的負荷は基質合成促進作用により基質の保持に不可欠の要素である半面、過大または過小な負荷は基質合成、分解の平衡破綻をきたし、椎間板変性促進の一つの素地になると推察された。
