癌細胞の骨転移形成に関わる破骨細胞形成因子とその作用機序

研究概要

臨床上で重要な問題となっている骨転移の機構を解析するために、まずヌードマウスの左心室内移植をした際に骨転移をおこすヒト乳癌細胞林並びにヒトメラノーマ細胞株を選択した。その結果、MDA-MB-231乳癌細胞株並びにA375Mメラノーマ細胞株が、移植後約6週間で骨吸収をともなった骨転移巣を形成することがわかった。これらのin vivoにおいて骨転移をおこす癌細胞の培養上清は、in vitroにおいて骨吸収を誘導しなかった。しかし、癌細胞の培養上清を骨芽細胞に添加して得た培養上清中には破骨細胞の形成を促進する因子が含まれており、この培養上清添加によりin vitroで骨吸収が誘導された。この破骨細胞形成因子は、IL-11であることが同定された。さらにIL-11に対する中和抗体によって骨吸収が抑制された。癌細胞はTGF-βを介して骨芽細胞からのIL-11産生を促進していた。よって、癌細胞が骨転移巣の拡大を行なう機構として、直接的に癌細胞が骨吸収を誘導しているというよりむしろ、間接的に骨芽細胞からの破骨細胞形成因子の分泌を促進することにより骨吸収をおこしていることが示唆された。今後、このIL-11を標的にした骨転移の治療法の開発を行なっていきたい。
