抗うつ薬による遺伝子発現制御機構の解明と抗うつ関連蛋白質のクローニング

研究概要	抗うつ薬は、治療効果を現わすまで数週間に渡る連続投与を必要としていることから、その作用機序に比較的長時間を要する遺伝子発現制御を介した機構が関与することが想像される。そこで、その点に着目した研究を行ない、imipramineが連投により脳内のセロトニン2Cサブタイプ受容体(5-HT2C)mRNA量を増大させることをin situ hybridization法を用いて見い出した。その作用様式について検討したところ、imipramineはヒト常用量にほぼ相当する5mg/kg、4日間経口投与においても5-HT2CmRNA増大作用を示し、20mg/kgまで濃度に依存して発現量が増大した。また20mg/kgを用いて投与日数による変化を検討したところ、投与4日目に発現量がピークに達し、その後少なくとも14日間はそのレベルを維持していた。Imipramine以外の抗うつ薬についても検討したところ、desipramine、mianserinが5-HT2CmRNA増大作用を示したが、nomifensineは無効であった。この結果は、抗うつ薬による5-HT2C遮断作用とよく一致した。5HT2CmRNA以外の遺伝子発現変化について、differential display法を用いて検討したところ、160bpの遺伝子がimipramine20mg/kg14日間投与により顕著に増大した。この遺伝子をDRF24と名付けたが、その分布をin situ hybridization法により検討したところ、vasolateral amygdaloid nucleusにおいてimipramine連投により発現が増大する可能性が示された。現在この遺伝子の全長のクローニングと生理機能の解明を行なっている。また、DRF24以外の同様の因子の検索も行ない、相互の関連性についても検討を行なう予定である。
