蛋白質燐酸化酵素PKNと相互作用する蛋白因子の同定

研究概要

PKNのアミノ末端をプローブとし、酵母two-hybrid法をもちいて、ヒト脳cDNAライブラリーをスクリーニングした結果、PKNと特異的に相互作用すると考えられる16種類のクローンを得た。そのうちの1種類はアミノ末端のactin結合領域を欠失したα-actininをコードしていた。まず初めに、α-actininとPKNとの相互作用に必要な領域を明らかにするため、それぞれの欠失変異体を用いたin vitro結合実験を行った。その結果、α-actininはPKNのアミノ末端領域と直接結合し、またその結合にはPKNのRhoA結合領域は不要であることがわかった。また、PKNは骨格筋型・非骨格筋型のα-actininの第三番目のspectrin-like repeatと結合し、さらに非骨格筋型のα-actininのEF-hand様領域とカルシウムイオン依存的にそして骨格筋型の同領域とはカルシウム非依存的に結合した。PKNとアミノ末端領域が欠失したタグ付きの変異体α-actininをCOS7細胞に共発現させ、その細胞抽出液よりタグに対する特異抗体をもちいて免疫沈降を行ったところ、この変異体α-actininとともにPKNが共沈することが明らかとなり、細胞内においてもPKNとα-actininが相互作用する可能性が示唆された。ウサギの網状赤血球抽出液を用いたin vitro翻訳系を利用して作製したactin結合領域を含む全長のα-actininは、in vitroにおいてPKNと弱い結合しか示さなかったが、アクチン細胞骨格系の調節に重要な働きをしているとされているphosphatidylinositol 4,5 bisphosphateを添加することにより、その結合活性は上昇した。これらのことから、PKNはα-actininと結合し、細胞骨格系に何らかの影響を与えている可能性が示唆される。
