膜アンカー型細胞増殖因子のジャクスタクリン/パラクリン変換の分子機構

研究概要

モンキー腎上皮由来Vero細胞にヒトproHB-EGFを発現した細胞株VeroH細胞を作成し、proHB-EGFのプロセッシングを免疫沈降法およびFACS分析により解析した。その結果、proHB-EGFの大部分は約1.5時間の半減期で細胞内に取り込まれ分解された。しかし、ホルボールエステル等によりC-キナーゼを活性化すると、proHB-EGFは約7分の半減期で分泌型HB-EGFへと効率よく変換された。このことは、HB-EGFのジャクスタクリン活性からパラクリン活性への変換がC-キナーゼを介する系路により制御されていることを示唆している。一方、CHO細胞に発現したヒト型proHB-EGFとマウス型proHB-EGFのジャクスタクリン活性に顕著な差異を見い出し、その原因をヒトーマウスキメラproHB-EGFを用いて解析した。その結果、proHB-EGFのN-末端プロセッシングがジャクスタクリン活性の発現に極めて重要であることを明らかにするとともに、このプロセッシングの担い手がエンドプロテアーゼの1つ、furinであることを示唆した
以上、本研究によりproHB-EGFのジャクスタクリン活性はエンドプロテアーゼ、furinによる末端プロセッシング及びC-キナーゼ系路を介するC-末端プロセッシングにより制御されていることを明らかにした。
