細胞接着による果粒膜細胞の分化調節機構

研究概要	卵巣の果粒膜細胞の分化によって発現するガングリオシドGM3とその糖鎖結合タンパク質の機能について、細胞接着の面から検討した。すなわち、(1)結合タンパク質と糖鎖との相互作用の解析、および(2)ガングリオシドの合成調節機構である。(1)について、糖鎖の誘導体(Alb-(SL)17)を放射性ヨウ素で標識し、培養細胞あるいはニトロセルロース膜上の糖鎖結合タンパク質との結合に及ぼす各種ムコ多糖類およびラット血清の影響を調べた。ムコ多糖類のうちヘパリンとヘパラン硫酸は他のムコ多糖類に比べて、糖鎖の誘導体の結合が強く抑制された。同様に、ラット血清も2%濃度でほぼ完全に糖鎖の誘導体の結合が抑制された。卵胞液中には高濃度のヘパラン硫酸が含まれること、通常の卵胞液には血清は含まれないが排卵時に血液が卵胞内に流れ込んでくることを考えると、この時期のガングリオシドと糖鎖結合タンパク質の相互作用は減弱していることが考えられる。(2)について、放射性ガラクトースで細胞を代謝的に標識して糖脂質の合成の後、糖脂質をTLCで分離して放射性ガングリオシドを測定する方法によって検討した。ガングリオシド合成はcAMPシグナルを介し、しかもエストロゲンに依存し、チロシンキナーゼ受容体ファミリーはこの合成を増幅したので、これら成長因子はガングリオシド合成のモデュレーターであることが示唆される。しかし、な何よりも分化した細胞のガングリオシド合成を最も上げる因子はLHである。ガングリオシドとその結合タンパク質の相互作用の減弱、および結合タンパク質の量の減少等を考えた場合、最終の細胞分化によってこのように増加するガングリオシドの合成は少なくとも細胞接着には関与しないことが示唆される。
