脳の微小循環動態の変化が神経組織への酸素供給に及ぼす影響

研究概要	脳の微小循環動態の変化にともなう脳組織への酸素供給状態の変化を定量的に評価することを目的とした。本年度は方法論の確立を目指した。1.測定装置は投下照明装置、実体顕微鏡、干渉フィルター、ビデオカメラ、画像処理装置で構成した。反射光を利用して、脳表面の微小血管網における赤血球の酸素飽和度ならびに赤血球流動挙動の解析を行った。2.微小血管網の血液の酸素飽和度は、ヘモグロビンの酸素化型と脱酸素化型の反射スペクトルの差を利用して求めた。本研究では、波長の異なる6枚の干渉フィルターを通して得られる顕微鏡画像をビデオカメラでとらえ、画像処理装置を用いてデジタル化し、コンピュータに取り込んだ。この画像データをKubelka-Munk理論に基づいて吸光度画像に変換した。6波長で得られた画像について重回帰分析を行って血液の酸素飽和度画像を求めた。ラット脳表面の微小血管網における血液の酸素飽和度の二次元画像の作成に成功した。3.方法の信頼性は酸素飽和度の異なる既知濃度の赤血球浮遊液を既知の光路長を持つ矩型ガラス管に封入して、顕微分光法で得られた結果と比較して確認された。4.ラットを用いて、麻酔下に総頚動脈を手術糸により牽引圧迫し、電磁血流計を用いて末梢側における血流量の減少を確認し、脳虚血条件設定を行った。しかし、脳表面の微小血管内における血流量の変化、赤血球の流速および赤血球の流動挙動については顕微鏡の低倍率および低解像度のため解析は困難であった。5.脳における微小循環動態と酸素放出動態の関連性を同時に解析できるようにするためには、ハード面で新しいシステムの開発が必要である。当面、本研究に対応できるシステムとしては、落射投光型顕微鏡に水浸対物レンズを用いるのが最適と考えられる。
