角化細胞増殖抑制因子の同定

研究概要

生体内における細胞数の調節に自己あるいは傍分泌性の増殖抑制因子が作用していると考えられている。皮膚角化細胞においては以前から動物実験によって、その存在が示唆されていたが、その実体はわかっていなかった。我々は角化細胞あるいは扁平上皮癌細胞の培養上清中に角化細胞の増殖抑制活性が分泌されることを見つけ、この活性の分泌は細胞密度と培養液のカルシウム濃度に依存することを報告した。これまでこの上清中に角化細胞の増殖抑制活性をもつ因子としてTGF-βSとIGFBP-6を同定していたが、上清中の主な活性分子はこれまでの方法では精製できなかった。本年度は、この分子の精製にさらなる改良が加えられ、角化細胞の増殖抑制活性を有する第3の分子を精製した。
1)角化細胞増殖抑制因子の精製
角化細胞増殖抑制活性を有する扁平上皮癌細胞TE-5の無血清培養上清を材料にして、BIO-REX70,QAEセファデックスのオープンカラム、メタノール/クロロホルム抽出、Superose12,Superdex peptideを用いたゲルろ過法、ならびにオクタデシルカラムを用いた逆相高速液体クロマトグラフィーによって精製した。
2)精製された分子の特性
精製された角化細胞増殖抑制因子は非蛋白性の小分子でHaCaT,Balb/MKなどの角化細胞株に増殖抑制活性を示したが線維芽細胞や血管内皮細胞には無効で扁平上皮癌細胞も耐性になっていた。現在、大量培養とマススペクトロメトリーなどによる構造解析を進めている。
