年齢推定に関する分子生物学的アプローチ

研究概要

成長の各ステージにおいて特異的に出現・消失する生体分子を指標とすれば年齢推定は用意であり、本研究ではこれら年齢依存性生体分子の検出・解析による個人の年齢推定を目的とした。我々はすでに数種類の年齢依存性生体分子を発見しており、本研究ではそのなかで17才以下の若年者尿中には存在するが、それ以上になると検出できなくなる酸性糖タンパク質(Ugl-Y)に着目した。同一個人より採取した尿より部分精製したUgl-Y標品を等電点電気泳動によって分離した後、Ugl-Yを膜フィルターに転写した。得られた転写物について、trypsinなどの数種類のタンパク質分解酵素処理を行いHPLCを利用して各種ペプチドを精製した。得られたペプチドのアミノ酸配列を気相ペプチドシークエンサーによって解析し、ugl-Yの一次構造(180アミノ酸残基)を決定した。その結果、その構造はfibronectinの一部にほぼ一致することが明らかとなった。次に、決定された一次構造に基づき作製したDNAフラグメントを利用したノーザンハイブリダイゼーションによって、fibronectinに対応するmRNA以外に1.5kb相当のmRNA分子種の存在が見出され、従ってUgl-Yはfibronectinのpost-translationalな産物ではなく、特有なmRNAより翻訳されているものと考えられた。今後の課題はUgl-Y特異的cDNAのクローニングであり、現在rapid amplification of cDNA end(RACE)法によるUgl-Y特異的cDNAの構築を計画している。
