心筋G蛋白質β3サブユニットの細胞質内局在の意味と共役蛋白質の検索

研究概要	当初網膜からクローニングされ、網膜に限局すると考えられたG蛋白質β3サブユニット(Gβ3)が、ラット心臓の細胞質に多く存在すること(Biochem Biophys Res Commun1995年)、さらにβ-アドレナリン受容体刺激により細胞質から細胞膜画分にTranslocationすること(Biochemical Pharmacology 1999年印刷中)を当研究室は報告した。ラット心臓におけるGβ3homologの存在が米国の研究グループにより示され(Gene 1995年)、さらに、ヒトGβ3のDeletion mutation(dominant gain-of-function)により、Na^+/H^+ Exchangerが活性化され、本態性高血圧症の病因となることがドイツの研究グループにより示唆された (Nature Genetics 1998年)。当研究室は、Gβ3に特異的な塩基配列をプローブとして、magnetic beadsを用いてヒト心臓cDNA Libraryから新しいGβ3Mutantを検索した。その結果、Gβ3Genomeのエクソン7上に8アミノ酸のDeletionをきたす新しいGβ3MUtantcDNAを同定した。また複数のヒト心筋RNAから、RT-PCRにて相応する2本のバンドを認めた。今回同定されたGβ3Deletionのドメインは、Gβγが様々なEffectorとInteractionするDomainで、このDeletionにより、細胞内情報伝達系に重大な異常が起きると予想される。心臓におけるGβ3の主要共役相手であるGγ5とのin vivoでの共発現により、細胞内情報伝達系への影響を検討していく計画である。
