膵臓のランゲルハンス島のβ細胞の形作りに関わるサイトカインの発生工学的解析

研究概要	(1)グルカゴンプロモーターによって、活性型TGF-β-cDNAをNODマウスの膵α細胞で発現させ、パラクラインとして膵β細胞に作用させたトランスジェニックマウス(Tg)では、TGF-βの免疫抑制作用によりIDDMの発症が抑制された。さらに、このTgのホモマウスを作成したところ、膵β細胞の発生が障害され、膵ラ氏島炎の機序を介さずにIDDMが発症した。また、膵ラ氏島炎を発症しているNODマウスから獲得したラ氏島を攻撃するTリンパ球を養子移入しても、TGF-βを発現しているTgでは対照に比べてラ氏島炎の発症が予防された。 (2)インスリンプロモーターによって、アクチビンの優性阻害受容体を膵β細胞で過剰発現し、アクチビンの情報伝達を阻害したところ、膵島の形成不全とグルコースに対するインスリン分泌不全が認められた。また、インスリンプロモーターによって、アクチビンの常時活性型受容体を膵β細胞で過剰発現したところ、やはり、膵島の形成不全が認められた。一方、正常型アクチビン受容体を膵β細胞で発現させたTgでは膵島に変化を認めなかった。したがって、アクチビンを初めとするTGF-βスーパーファミリーの適正な情報伝達が膵島の発生に重要であることが示唆された。 (3)グルカゴンプロモーターによって、regIを膵α細胞で発現させたTgでは膵ラ氏島の巨大化、糖尿病の発症、膵腫瘍の発生などが認められた。したがって、regIは膵島細胞の増殖や機能に重要な役割を果たすことが示唆された。
