腎臓における新しい細胞間情報伝達蛋白質のクローニング-シグナルシークエンストラップ法を用いて-

研究概要	1.シグナルシークエンストラップ(SST)法によるマウス腎臓cDNAライブラリーのスクリーニング:8週齢雄性BALB/cマウス腎臓より抽出したmRNAから2本鎖cDNAを合成し、シグナルシークエンス部分を欠失させたIL-2受容体α鎖(Tac)cDNAの5'側に連結した。これをプラスミドベクターpcDL-SRαを用いてCOS-7細胞に発現させたところ、シグナルシークエンスが挿入された場合にのみ、融合蛋白質が細胞膜表面に到達し、抗Tac抗体による蛍光染色にて認識された。5000クローンをスクリーニングすることにより、25個の陽性クローンが得られた。塩基配列を決定した結果、このうち13個は既知蛋白質として、分泌蛋白質(ウログアニリン)、膜結合型受容体(シグナルシークエンス受容体)、接着分子(Kspカドヘリン)、チャンネル(アクアポリン2)、トランスポーター(Na依存症リン酸トランスポーター)が得られた。以上より、腎臓に発現する分泌型及び膜結合型蛋白質のクローニング法としてのSST法の有用性が証明された。 2.新しい腎臓由来アスパルティックプロテアーゼ様蛋白質のクローニング:SST法にてマウス腎臓より得られた67アミノ酸からなる未知のポリペプチド断片をコードするクローン(E3A5)の塩基配列をもとに3'RACEを施行し、1.2kbの単一バンドを得た。cDNA全長の塩基配列の構造決定により、419アミノ酸からなるアスパルティックプロテアーゼ様蛋白質であることが明らかとなった。本蛋白質はカテプシンD、カテプシンE、レニンと47%、46%、42%の相同性を有していた。ノーザンプロット解析によりmRNAは、サイズが1.6kbで、腎臓に最も高濃度に発現しており、肺、脾臓、脂肪組織にも検出された。In situハイブリダイゼーション法により、その腎内局在を検討したところ、近位直尿細管に限局した強い遺伝子発現を認め、近位曲尿細管にも弱い発現を認めた。本プロテアーゼ様蛋白質の腎臓における遺伝子発現は近位尿細管に特異的であり、再吸収された蛋白質をライソゾーム内で分解する新しいカテプシンと考えられた。
