研究概要

(1)カブトガニ軟骨由来のコンドロイチン硫酸Kから新規の四糖〜七糖、およびイカ軟骨由来のコンドロイチン硫酸Eから新規の四糖をそれぞれ単離し、完全一次構造を決定することに成功した。それらはグルクロン酸-3-硫酸をもつものや、フコースの結合した珍しい構造を含んでいた。それらを基質として、構造解析に必要なコンドロイチナーゼとヒアルロニダーゼの基質特異性を明かにした(論文2報)。
(2)サメ軟骨由来のコンドロイチン硫酸Dから新規の八糖を単離し、完全一次構造を決定することに成功するとともに、神経突起伸長促進活性を示すことを明らかにした(論文)。
(3)コンドロイチン硫酸の生合成に関わるβ-GalNAc転移酵素とβ-GlcA転移酵素の基質特異性を調べ、受容体基質の硫酸化パターンによって酵素による糖転移の活性が大きく変動することを明らかにした(論文2報)。
(4)ブタ小腸由来のヘパリンから新規の四糖、五糖、六糖を単離し、完全一次構造を決定することに成功した(論文)。
(5)アフリカツメガエルの尾芽胚から調製したヘパラン硫酸鎖が細胞増殖因子bFGFや細胞の分化誘導調節因子であるFollistatinと特異的な相互作用をすることを見い出し、その分子認識の性質のいくつかの特徴を解明した(論文)。
