研究概要

本研究は、脳における情報処理や神経伝達機構などの、生物の持つ優れた機能を持ち、且つ工学的に応用可能な全く新しい「生体機能分子」の「設計・合成」を行うことを目的とする。
生体機能分子を設計・合成するために、有機合成化学的アプローチと蛋白質化学的アプローチの2つの方針をとる。有機合成的アプローチとしては、モレキュラーインプリンティング法による分子認識部位の構築と触媒能の導入を行う。タンパク質工学的アプローチとしては、タンパク質の化学修飾による電子伝達効率の促進と、天然メディエーター分子の化学修飾による酸化還元特性の改変を行う。
平成8年度は具体的に以下の2つの研究目的とした。
1.モレキュラーインプリンティング法により創製した生体機能分子を、高性能分離素材、センサー素材、エレクトロニクス素材、メカニカル素材などに応用する。
2.酵素-人工メディエーターハイブリッド分子を創製し、バイオチップへの応用を図る。
これらを目的に対して以下の成果が得られた。
1.モレキュラーインプリンティング法により分子認識ポリマーを合成し、更にこれを化学修飾することにより、ポリマーの分子認識機能を向上させた。
2.モレキュラーインプリンティング法により合成した分子認識ポリマーの分子認識を、色素を用いて検出を簡素化し、センサーへの応用の可能性を示した。
3.蛋白質と合成高分子や電子メディエーターをハイブリッドさせて電子伝達機能を改変し、特性を評価した。
更に従来の方法と全く異なる生体機能分子の創製法である「バーチャルエボルーション」という手法の可能性を確認することができた。
