系統解析によるマメ-マメゾウムシ-寄生蜂群集の共進化的な生成過程の解析

研究概要	マメ科植物-マメゾウムシ科-寄生蜂群集を対象に、構成種の「被食-捕食」関係がどのように共進化的に生成したのかを、被食者-捕食者双方の分子系統樹の対応関係から分析を試みた。今年度の種子の調査は、アジアではタイ(藤井、徳永が1回、津田グループが2回)、ミャンマー(藤井、徳永)、西アフリカではトーゴとブキナ・ファソ(ツール大学のMonge、Huignard)、オセアニアではオーストラリア(徳永)、新大陸では米国アリゾナ(嶋田グループが2回)にて行った。採集したマメ科種子は、嶋田あるいはHuignardの実験室(ともに植物防疫の輸入許可付き)に持ち込み、羽化したマメゾウムシと寄生蜂の寄主を記録して標本にし、さらに生きた成虫をアルコール漬けにしてDNA分析試料とした。マメゾウムシの標本は協力者のC.D.Johnson(北アリゾナ大学・生物学教室・教授)へ、寄生蜂は協力者のCasas(ツール大学・農業生物実験研究所・教授)へ送り、それぞれ種を同定した。虫のアルコール漬け試料は、嶋田と津田がCox-I・Cox-II・ND5などのmtDNA領域を用いて、PCR-RFLP法とシーケンサのDNA情報により系統樹の作成を開始した。また、タイ、ミャンマーから採集されたヨツモンマメゾウムシ、アズキゾウムシに関しては、筑波大学において累代的に飼育可能な系統が確立された。
