研究概要

蛋白質では構造の複雑性を反映して、さまざまな階層で揺らぎが生じることが予想される。このような揺らぎが協同的に働いてより大きな階層の揺らぎを作りだし、最終的には蛋白質の機能を生み出すと考えられる。また、機能は折り畳まれて初めて発現されるので、折り畳みに伴いこのような揺らぎは変化するであろう。然るに、このような揺らぎ(蛋白質動力学)についての理論的研究は進展しているが、実験的研究はほとんどなされてこなかった。本国際共同研究は、蛋白質動力学の実験的研究の展開及び理論研究との融合をめざして組織された。
黄色ブドウ球菌の核酸分解酵素を用いて、室温及び25Kの中性子非弾性散乱スペクトルを広いエネルギー範囲で観測した。蛋白質の非弾性散乱スペクトルとしては、これまで報告されている中で、最高精度のデータを得ることができた。得られたスペクトルを、基準振動解析の結果と比較した。定性的には両者の一致は良く、観測されたピークを帰属することが可能であった。理論計算の結果が、実験的に検証された初めての例である。しかし、ピーク位置のシフトが見られるなど一致は完全ではなく、特に強度の不一致が著しかった。この結果は、理論計算に用いられているポテンシャル関数に改善の余地のあることを示している。
同蛋白質及び変異体について、中性子準弾性散乱測定を行った。変異体は生理的溶媒条件下でコンパクトな変性状態にある。準弾性散乱測定の結果、非調和的な運動は変性状態の方が顕著であるが、低エネルギーの協同的な調和的な運動は折り畳まれた状態の方がより強く観測された。準弾性散乱から評価された平均自乗変位の折り畳みに伴う変化量は、MDシミュレーションから推定される値に近かった。以上の結果は、機能動力学の密接な関係を示唆している。
