蛋白質の立体構造安定性マップ作製のための基盤研究

研究概要	最近、ヒトゲノムを始め多くの遺伝子配列が幾何級数的に日々明らかにされている。しかし、その膨大な遺伝子配列から蛋白質の機能・構造を推定することはほとんど不可能である。あるアミノ酸配列に適合する既知の立体構造を探して未知蛋白質の立体構造を予測するいわゆる3D-1D法が有力視されている。しかしこの予測に用いられている評価関数も経験的なラフなものである。それは、蛋白質の個々のアミノ酸残基が立体構造上の特性との関連で、蛋白質立体構造の安定性にどのように寄与しているか定量的に明らかにされていないことに原因している。本研究計画では、「蛋白質の個々のアミノ酸残基が立体構造上の特性との関連で、蛋白質立体構造の安定性にどのように寄与するかを明らかにすること」、つまり、「蛋白質立体構造安定性マップ」の作製を行うための基本的な問題点を明らかにし、マップ作製が可能となるように基盤を整備することを目的としている。この解決手段の一つとして、西川らは全ての可能な一残基置換の安定性を一挙に表現する「変異型プロフィール」のテーブルを作成している。本研究では、ヒト・リゾチーム(油谷)とユビキチン(Makhataze)の種々の構造上の部位で、置換残基の特性と安定性との関連を明らかにする。ヒト・リゾチームにおいて、違った部位での同じ性質のアミノ酸置換体を系統的に作成し、それらの安定性の変化を高精度の示差走査熱量計で測定し、構造変化をX線結晶解析で求めた。分子内部、分子表面、αヘリックス、β構造、ループなどに位置している変異型約20種について調べた。それらの結果と「変異型プロフィール」からの予測値との差異を検討中である。Makhatazeはユビキチンの特定部位に焦点をあてて、一つの構造の特徴を徹底的に研究しているが、彼とは熱測定の解析法まで踏み込み得られたデータの解析と検討を共同で行っている。
