ヒト7番染色体の不死化抑制遺伝子の単離

研究概要

酵母TAR(transformation associated recombination)YAC/BACベクターを用いて、マウス放射線雑種細胞S10-3-2(ヒトDNAを2-3Mb含む)からヒトDNAを300個のBACクローンとして回収した。YACクローンを大腸菌内でBACに変換し、不死化細胞株Met5Aへの遺伝子導入によって、その分裂を停止するBACクローンを数個単離した。しかし、分裂停止という負の選択では、目的遺伝子以外でも分裂停止を誘導することが起こることが判明し、BACクローンの解析はまだ完了していない。一方、ヒト7番染色体を移入し、分裂が停止したMet5A細胞クローンから多数のリバータントを分離した。これらリバータントについてSTSマーカーを用いたPCR解析を行い、移入されたヒト7番染色体上の欠失部位のマッピングをを行った。共通の欠失部位が存在すれば、そこに目的遺伝子が乗っていることが示唆されるが、そのような領域はまだ特定されていない。一方、7番染色体は、D群不死化細胞のテロメラーゼ活性の抑制およびテロメアの短縮を誘導することを示した。7番染色体はテロメラーゼ活性をもたいない不死化細胞株のテロメアの短縮も誘導するので、細胞老化の誘導を介してテロメア維持機構を負に制御することが示唆された。
