蛍光新技術を用いた蛋白-蛋白および蛋白-DNA相互作用の動的解析

研究概要	蛍光を含む生物物理的手法で、大腸菌RNAポリメラーゼの転写開始を司るσ^<70>サブユニットの転写における機能を研究した。 1.他サブュニツトとの接触には、他細菌類と類似するσ^<70>の4つの領域が役割を持ち、相互作用部位の広い特異なタンパク質-タンパク質相互作用を持つことを、ヒドロキシルラジカルフットプリント法で明らかにした。 2.他細菌類には無いσ^<70>のNC領域にクラゲ蛍光タンパク質であるGFPを導入した活性のあるσ^<70>を作製した。 3.NC領域の機能を確認するために、これを欠失したσ^<70>(delNC)を作製し、その機能が、σ^<70>の溶解度を向上させ、オリゴマー形成を阻止することが明らかになつた。領域2と3にCys残基を一つ導入した変異体delNCを、4種作製し、細胞内での活性測定を行い、3種は完全にな活性、一種は50%程度を示した。この5種のdelNCを、in vitroで活性を測定すると、プロモーターに依存して活性が変化し、NC領域は、反応にも寄与していることが明らかになった。 4.レーザーフォトリシスに適したラジカル生成試薬を合成し、変異体delNCのCys残基を修飾し他サブユニツトやDNAとの相互作用を研究する準備を行った。 5.ωを欠くコア酵素は、σが結合せず活性はなく、GroELが強固に結合していることを発見した。今まで未知であったωの機能が、ポリメラーゼを活性型のコンフォーメーションに変換する機能を持つことを明らかにした。GroESとの相同性のある領域を発見、ωがGroELのATPase活性をGroESと同様に阻害することが判明、GroESの役割、GroELの放出、を担うことが示唆され、built-in shaperon analogueという新規のタンパク質であることが示唆された。
