感染防御におけるIL-12の役割

研究概要

我々は、昨年まで赤内型P.bergheiの強毒・弱毒株感染におけるIL-12の感染防御と病態形成への関与という二つの役割を明らかにしてきた。今年度は、昨年までの結果をさらに発展させ、赤内型弱毒株P.bergheiXAT感染におけるIL-12およびIFN一γの下流に位置する感染防御のエフェクター機構として、NK細胞とNOの関与について検討した。まず、強毒・弱毒感染でそれらを比較したところ、いずれもその活性や産生量が強毒株感染に比べ弱毒株感染で有意に高く感染防御との正の相関性が得られた。しかし、抗NK1.1抗体をマウスに投与しNK細胞を除去し、NK活性がほとんど無くなってもParasitemia変化や脾細胞からのIFN-γ産生には有意な差は見られず、感染防御への関与は低いことが明らかになった。さらにiNOSノックアウトマウス(KO)に感染させても、Parasitemia変化やIFN-γ産生には有意な差は見らなかったが、このマウスヘ抗INF-γ抗体を投与するとParasitemiaが上がり続け死亡した。またCD4KOマウスでは、脾細胞からのIFN-γ産生が著明に低下し、感染赤血球を除去できずに死亡した。これらより、NO産生の感染防御への関与は低くCD4^+T細胞から産生されるIFN-γを介した他の機構の重要性が明らかになった。この機構として感染初期と後期のそれぞれに働く二つの機構を考えている。前期は、非特異的防御機構、後期は抗体を介した食細胞の貪食やADCCによる機構を考え現在検討中である。一方、肝臓型マラリア感染に対するワクチン開発を目的に、P.yoeliiおよびP.falciparumのCS蛋白を有する種々の組み換えウイルスを作製しマウスに免疫し誘導されるcs蛋自に特異的CD8^+T細胞数を測定しその防御免疫誘導能を比較したところ、アデノウイルス単独投与およびインフルエンザウイルス投与の後ワクシニアウイルスでブーストをかける方法により、防御免疫がより強く誘導されることが明らかになった。
