SAチャネルの分子生物学と生理学の展開に関する共同研究

研究概要	成果報告 SAチャネルは、その分子実体と生理的役割の解明が待ち望まれているイオンチャネルの一つである。本計画は、我々のこれまでの共同研究の成果(特異的ブロッカーの発見、遺伝子クローニングの進行)に基づいて、その基礎的・応用的研究を展開することである。具体的には、1)SAチャネルのcDNAを組み込んだ細胞系の開発とその応用、2)SAチャネル特異的ブロッカーの開発とその応用の2大目標である。これに加えて、生細胞への定量的機械刺激法、および単一SAチャネル電流の効率的解析法の開発を行う。研究は、分担者がそれぞれの研究室で連絡をとりながら研究を進め、適宜材料を持ち寄って共同実験、研究打ち合わせを行うというスタイルで進めた。その結果、1)SAチャネルの候補遺伝子であるmec-4をMDCK細胞(cell line)に組み込んだ細胞系をつくることができた。しかし、組み込み後の細胞寿命が短く、現在この解決法を探っている。2)CHO細胞に、ある遺伝子(未公表)を組み込み、これがSAチャネル活性を持つことが分かった。現在その安定化と解析を進めている。3)顕微鏡下で定量的かつ生理的一軸伸展刺激を与えられる装置を開発した(以上曽我部担当分)。4)ブロッカーについては、一旦精製されたがやや再現性に欠けるため、現在その条件確立に向けて検討中である。5)原子間力顕微鏡(AFM)のプローブを用いた定量的機械刺激法を開発した。6)MLF法に基づいたチャネルキネティクスの効率的解析法を開発し、SAチャネルの短時間記録の解析に成功した(以上Sachs担当分)。曽我部らは計3回の派遣で、ブロッカー精製の助言と共同実験を行った。またSachsらが開発したAFM刺激法とチャネル解析法を修得するとともに、今後の研究打ち合わせを行った。
