研究概要	メラノーマ特異抗原で細胞障害性T細胞(CTL)を惹起できることが期待されるgpl00の遺伝子をマウスに発現させ、宿主の免疫学的反応の解析と、実際に担癌マウスでの腫瘍抑制効果を検討した。ヒトgp100遺伝子をCMVプロモーターの下流につなぎHVJ-liposomeによってC57BL/6マウスの骨格筋に2週間隔で2回注入すると、gp100に対する抗体が産生されていることが免疫沈降法により確認された。マウスメラノーマ細胞株B16を攻撃できるCTLがマウス牌臓中に産生されていることも明らかになった。次に、gp100発現ベクターで2週間隔で2回免疫したC57BL/6マウスの足底に2500個のB16細胞を接種した。gp100を用いなかったマウスでは約3週間で腫瘍の形成を認め、腫瘍注入後70日以内に全例死亡した。gp100で免疫したマウスにおいて腫瘍の形成が遅延し、生存の長期化がえられたが、130日後に全例死亡した。さらに効果的な免疫を得るため、一過性に強い発現をおこせるベクターとしてセムリキ森林ウィルスベクターを用いて試験管内で合成したmRNAをHVJ-11posomeで組織で発現させることにした。またその注入部位として骨格筋ではなく抗原提示細胞が豊富と考えられる牌臓を選択した。同様の免疫実験を行い、抗体、CTLに誘発を確認し、腫瘍形成に対する効果をみたところ、gp100mRNAで免疫したもののみ、160日後においても6匹中2匹が生存し、腫瘍の再投与でも腫瘍形成を認めなかった。以上のことは、gp100遺伝子がメラノーマワクチンとしてはたらくこと、gp100のRNAはより安全なワクチンとして効果が期待できること、投与部位を筋肉ではなく抗原提示細胞が豊富と考えられる組織(牌臓やリンパ節)を選択する必要があることを示唆している。
